The six 9D7 Fab CDR loops interact with a discontinuous IL-10M1 epitope comprised of residues 71–83 and 125–137. 9D7 non-competitively antagonises IL-10R1 binding.

Preparation:
Rats are immunised with synthetic oligopeptide (containing an N-terminal cysteine)-carrier protein conjugates. IL-10 was conjugated to JES3. The mouse non-secreting myeloma fusion partner P3X63-AG8.653 is fused to immunised rat splenocytes to produce heterohybridomas secreting rat antibody. Bioassays were used to identify neutralising mAbs, and immunochemical screens were used to identify mAbs that bind the cytokine.

Target Information

IL-10 encodes a protein that acts as a cytokine and is primarily produced by monocytes, with some production by lymphocytes. This cytokine has various effects on immunoregulation and inflammation, including down-regulating the expression of Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. Additionally, IL-10 can block NF-kappa B activity and is involved in the regulation of the JAK-STAT signaling pathway. Knockout studies in mice suggest that this cytokine is an essential immunoregulator in the intestinal tract. Mutations in this gene have been associated with increased susceptibility to HIV-1 infection and rheumatoid arthritis. Diseases associated with IL10 include Graft-Versus-Host Disease and Human Immunodeficiency Virus Type 1.
