Product Specific Information

The 236A/E7 antibody reacts with human Foxp3 protein (FORKHEAD BOX P3, SCURFIN, JM2). Foxp3 (49–55 kDa) is a transcriptional regulator from the forkhead/winged-helix family, identified as the defective gene in scurfy mice. High Foxp3 expression is characteristic of CD4+CD25+ regulatory T cells (Treg), and ectopic expression in CD4+CD25– cells induces a Treg phenotype.

Using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523), intracellular staining of human PBMCs shows specific staining of CD4+CD25bright populations.

The epitope recognized by 236A/E7 differs from that of PCH101 (Product #72-5776-40). This antibody also recognizes rhesus macaque, sooty mangabey, and cynomolgus macaque.

Applications Tested:

  • Immunoblotting and IHC of formalin-fixed paraffin-embedded tissue using antigen retrieval (low or high pH).
  • Recommended concentration: 1–10 µg/mL for WB; ≤5 µg/mL for IHC.
  • Titrate for optimal assay performance.

Target Information

FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis. It is stably expressed in CD25+CD4+ regulatory T cells, at low levels in CD4+CD25– cells, and absent in CD4–CD8+ T cells. FOXP3 acts as a master regulatory gene and marker of Treg cells. Defects cause the scurfy phenotype in mice and IPEX syndrome in humans (X-linked autoimmunity-allergic dysregulation). Transcript variants encoding different isoforms have been identified. In human breast and colon cancer, FOXP3 expression is regulated by p53 in response to DNA damage.
