Target Information
MLCK (also known as MYLK3) is a member of the myosin regulatory light chain kinase family that facilitates muscle contraction. It phosphorylates specific serine residues on myosin light chains (MLC), promoting interaction with actin filaments and enabling smooth muscle contraction.
MLCK plays a central role in vascular permeability and tissue edema formation. In the nervous system, it contributes to astrocytic process growth and neurotransmitter release. It is also involved in fibroblast apoptosis signaling.
Both smooth and non-muscle isozymes are expressed in various adult and fetal tissues. The non-muscle isoform 2 is the dominant splice variant, while the Telokin isoform binds calmodulin and is widely expressed.
MLCK contains one fibronectin type III domain and nine immunoglobulin-like C2-type domains and is likely down-regulated by phosphorylation.
