Target Information

MLCK (MLCK2), also known as MYLK3, is a myosin regulatory light chain kinase that facilitates muscle contraction. As a member of the Ser/Thr protein kinase family, MLCK phosphorylates a specific serine in the N-terminus of myosin light chains (MLC), promoting interaction with actin filaments. It plays a central role in vascular permeability and tissue edema formation. In the nervous system, MLCK regulates astrocytic process growth and transmitter release at synapses, and participates in fibroblast apoptosis signaling. Both smooth muscle and non-muscle isoforms are widely expressed, with non-muscle isoform 2 being predominant. The Telokin isoform binds calmodulin and is found in various tissues. MLCK activity is likely down-regulated by phosphorylation and the protein includes one fibronectin type III domain and nine immunoglobulin-like C2-type domains.
