Description:
The 10H11.E12 monoclonal antibody recognizes human, mouse, and rat ATM protein phosphorylated on serine 1981. ATM, a member of the PI3 kinase family, coordinates cell cycle arrest and DNA repair in response to DNA damage and oxidative stress. Upon activation by DNA breaks, ATM monomers form through autophosphorylation and localize to DNA double-strand break sites to initiate repair. Dysfunction of ATM leads to ataxia-telangiectasia (AT), characterized by neurodegeneration, immunodeficiency, and cancer predisposition.
Applications Reported:
Validated for intracellular staining followed by flow cytometric analysis.
Applications Tested:
Pre-titrated and tested for intracellular staining of normal human peripheral blood cells. Recommended usage is 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Fluorochrome Information:
eFluor® 660 replaces Alexa Fluor® 647. Excitation: 633–647 nm; Emission: 668 nm; Laser: Red.

Target Information
Ataxia-telangiectasia Mutated (ATM) is a PI3/PI4 kinase family protein involved in DNA double-strand break repair. Mutations in ATM cause ataxia-telangiectasia, a recessive disorder marked by neurodegeneration, immune deficiency, and cancer susceptibility. ATM phosphorylates key regulators such as p53, Mdm2, BRCA1, and SMC1, and is activated by ionizing radiation to regulate cell cycle, apoptosis, and DNA repair pathways.
