Product Specific Information

This SC1T2M3 monoclonal antibody recognizes human and mouse Src tyrosine kinase when phosphorylated on tyrosine 418 (Y418).
Autophosphorylation of Src at Y418 in the catalytic domain is required for full catalytic activity. Src is a non-receptor tyrosine kinase involved in signal transduction in numerous biological systems and is activated following engagement of immune receptors, integrins, receptor tyrosine kinases, G protein-coupled receptors, and cytokine receptors.
Aberrant Src activity has been implicated in various cancers. Due to sequence homology surrounding Src Y418, this clone may cross-react with other Src family kinases including Lck, Fyn, and Lyn.

Specificity was determined by ELISA, flow cytometry, and western blotting.

Applications Reported:
Intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by intracellular staining of stimulated human peripheral blood cells. Use at 5 µL (0.03 µg) per test (defined as antibody amount for 100 µL final volume). Suitable for 10⁵–10⁸ cells/test.

Target Information

SRC is a cytoplasmic tyrosine kinase and proto-oncogene involved in embryonic development and cell growth regulation. It associates with various cell surface receptors to mediate signal transduction and plays roles in cytoskeletal organization, cell cycle regulation, and apoptosis.
Phosphorylation at Tyr416 activates SRC, while Tyr527 phosphorylation inhibits it. Mutations in SRC may contribute to malignant progression of colon cancer. The SRC gene is located on chromosome 20, with two transcript variants encoding the same protein.
