Product Specific Information
This antibody has been negatively preadsorbed using a non-phosphopeptide corresponding to the phosphorylation site to remove reactivity with non-phosphorylated Tau. The final antibody is purified by affinity chromatography using a Tau-derived peptide phosphorylated at serine 396.
Target Information
Tau is a neuronal microtubule-associated protein primarily localized in axons. It promotes tubulin polymerization and stabilizes microtubules. The C-terminus binds axonal microtubules, while the N-terminus binds neural plasma membrane components, acting as a linker between them. Tau localization defines axonal polarity and contributes to cytoskeletal plasticity and stability. Hyper-phosphorylated Tau forms paired helical filaments (PHF), a major component of neurofibrillary tangles in Alzheimer’s disease (AD) brains. This modification impairs Tau’s microtubule binding, leading to neuronal degeneration. Phosphorylation is mediated by kinases including GSK-3β, PKA, cdk5, and casein kinase II. Abnormal Tau phosphorylation is implicated in AD, Pick’s disease, frontotemporal dementia, corticobasal degeneration, and progressive supranuclear palsy.
