Target Information
Heterotrimeric G proteins are membrane-bound GTPases associated with 7-transmembrane receptors. Each G protein consists of alpha, beta, and gamma subunits and remains bound to GDP in the “off” state. Upon ligand-receptor binding, the G protein detaches, switching to the “on” state and allowing Galpha to activate second messenger signaling cascades. Galphas and Galphai directly interact with adenylyl cyclase to stimulate or inhibit its activity, resulting in increased or decreased cAMP levels, respectively.
