Product Specific Information
The eBio124-1D1 monoclonal antibody reacts with human CD7, also known as gp40 or Leu9.
CD7, a 40 kDa receptor, is part of the immunoglobulin gene superfamily. The N-terminal region (aa1–107) is homologous to Ig kappa light chains, while the C-terminal extracellular domain is proline-rich, forming a stalk for the Ig domain.
CD7 is expressed on most immature and mature T lymphocytes, T cell leukemias, natural killer cells, a subset of normal B cells, and malignant B cells.
Cross-linking of CD7 enhances T cell and NK cell activation, influencing calcium flux, adhesion molecule expression, cytokine secretion, and proliferation. CD7 directly associates with phosphoinositol 3'-kinase, inducing D-3 phosphoinositide production and tyrosine phosphorylation.
A clonogenic subpopulation of human CD34(+) CD38(-) cord blood cells expressing CD7 has lymphopoietic potential, generating B cells, NK cells, and dendritic cells but not myeloid or erythroid cells. CD7(+) phenotype distinguishes primitive lymphoid progenitors from pluripotent stem cells.
CD7 may cooperate with CD28 during Treg function, as CD28/CD7-deficient mice show reduced Treg numbers and suppressive activity.

Applications Tested
- Pre-titrated and tested by flow cytometry on normal human peripheral blood cells.
- Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume, 10^5–10^8 cells/test).
Handling and Storage
- Light Sensitivity: Protect from light (photo-induced oxidation).
- Fixation: Samples can be stored up to 3 days in the dark at 4°C using IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333).
- Excitation: 488–561 nm
