Product Specific Information
This antibody detects endogenous PAK1/PAK2 protein at a molecular weight of approximately 60–70 kDa.
Purity is greater than 95% as determined by SDS-PAGE.
Target Information
PAK (p21-activated kinases) are serine/threonine kinases that include three main isoforms: alphaPAK (p68), betaPAK (p65), and gammaPAK (p62). These isoforms share high sequence homology with the S. cerevisiae kinase Ste20, which is involved in pheromone signaling.
PAK proteins interact specifically with Rac1 and Cdc42 in their active GTP-bound state, inhibiting their intrinsic GTPase activity and leading to autophosphorylation.
GammaPAK has eight autophosphorylation sites, including Ser19, Ser141, and Thr402, and phosphorylation of Ser141 and Thr402 correlates with activation. Once phosphorylated, PAK isoforms dissociate from Rac/Cdc42 complexes to interact with downstream substrates.
