Product Specific Information

The B-Ly7 monoclonal antibody reacts with human CD103, the alpha E integrin. CD103 non-covalently associates with integrin beta 7 and is expressed mainly on intraepithelial lymphocytes and a small subset of peripheral lymphocytes. It is also expressed in hairy cell leukemia (HCL) and some chronic B cell lymphocytic leukemias. In vitro stimulation of human T cells with mitogens induces CD103 upregulation. E-cadherin binds to CD103 and mediates lymphocyte homing to intestinal epithelium.

Each product contains:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of 2–3 day PHA-activated human peripheral blood cells. Use at 5 µL (0.4 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Recommended: Invitrogen LIVE/DEAD Fixable Dead Cell stains.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
• Add 5 µL per sample (regardless of NovaFluor count) before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, employing novel nucleic acid dye structures for engineered fluorescent signatures with reduced spillover and spread.

Target Information

CD103 (ITGAE, Integrin alpha E) is expressed on mucosa-associated T lymphocytes and activated cells. It comprises a 150 kDa alpha chain and a 120 kDa beta chain. Expressed by 0.5–2% of resting lymphocytes in peripheral blood and lymphoid organs. Stains a few bone marrow cells and rarely peripheral lymphocytes.

CD103 belongs to the integrin family—heterodimeric membrane proteins with alpha and beta chains. ITGAE encodes an I-domain-containing alpha integrin that undergoes extracellular post-translational cleavage, forming disulfide-linked heavy and light chains. Together with beta 7 integrin, CD103 forms the E-cadherin binding integrin (human mucosal lymphocyte-1 antigen). Preferentially expressed in intestinal intraepithelial lymphocytes (IEL), it aids adhesion and activation.
