Product Specific Information

This H5C6 monoclonal antibody reacts with human CD63, a type III member of the tetraspanin family of transmembrane proteins. CD63 is expressed intracellularly on lysosomes, endosomes, and granules of resting platelets and basophils. Cell surface expression of CD63 can be detected on activated basophils and platelets, monocytes, macrophages, and granulocytes. It is also expressed on endothelial cells, fibroblasts, and smooth muscle cells. CD63 associates with integrins (VLA-3 and VLA-6) and TIMP-1 to mediate allergic responses.

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Use 5 µL (1.0 µg) per test in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.

Important Usage Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus per sample or per 100 µL single-color control.

NovaFluor conjugates utilize Phiton™ technology, which employs nucleic acid dye structures for engineered fluorescent signatures with reduced spillover and spread.

Target Information

CD63 (LAMP-3) is a glycoprotein of the tetraspanin family, present in late endosomes, lysosomes, and secretory vesicles. It appears on the plasma membrane following cell activation and serves as a basophil activation marker. CD63 interacts with integrins, affects phagocytosis and cell migration, and participates in H/K-ATPase trafficking and ROMK1 channel regulation. It also functions as a T-cell co-stimulation molecule. CD63 expression can indicate carcinoma prognosis and is associated with melanoma and Hermansky-Pudlak Syndrome.
