Product Specific Information

This H5C6 monoclonal antibody reacts with human CD63, a type III member of the tetraspanin family of transmembrane proteins. CD63 is expressed intracellularly on lysosomes, endosomes, and granules of resting platelets and basophils, and on the cell surface of activated basophils, platelets, monocytes, macrophages, and granulocytes. It is also found on endothelial cells, fibroblasts, and smooth muscle cells.

CD63 associates with integrins (VLA-3 and VLA-6) and TIMP-1 to mediate allergic responses.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells; use 5 µL (0.8 µg) per test (final volume 100 µL; 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures designed to minimize spillover and spread.

Target Information

CD63 (LAMP-3) is a glycoprotein of the tetraspanin family found in late endosomes, lysosomes, and secretory vesicles. It is also expressed on the plasma membrane following activation. CD63 serves as a basophil activation marker and interacts with integrins, influencing phagocytosis and cell migration. It is involved in H/K-ATPase trafficking and ROMK1 channel regulation, and acts as a T-cell costimulation molecule.

CD63 expression can serve as a prognostic marker in early-stage carcinomas. It is expressed on activated platelets, monocytes, macrophages, and weakly on granulocytes, B, and T cells. CD63 is identical to melanoma antigen ME491 and platelet antigen PTLGP40.
Diseases associated with CD63 dysfunction include melanoma and Hermansky-Pudlak Syndrome.
