Product Specific Information

The B-Ly7 monoclonal antibody reacts with human CD103, the alpha E integrin. CD103 non-covalently associates with integrin beta 7 and is mainly expressed on intraepithelial lymphocytes and a small subset of peripheral lymphocytes. It is also expressed by hairy cell leukemia (HCL) and some chronic B cell lymphocytic leukemias.
E-cadherin binds to CD103 and mediates homing of lymphocytes to the intestinal epithelium.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and tested with 2–3 day PHA-activated human peripheral blood cells at 5 µL (0.4 µg) per test.
A test is defined as the amount of antibody staining a 100 µL cell sample (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
• Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.

Target Information

CD103 (ITGAE, Integrin alpha E) is expressed on mucosa-associated T lymphocytes and activated cells, comprising a 150 kDa alpha chain and a 120 kDa beta chain.
It is found on 0.5–2% of resting lymphocytes in peripheral blood and lymphoid organs, and occasionally on bone marrow cells.
CD103 forms the E-cadherin binding integrin with beta 7 and is preferentially expressed in intestinal intraepithelial lymphocytes (IEL).
Associated diseases include hairy cell leukemia and splenic marginal zone lymphoma.
