Target Information
Post Synaptic Density 95 kDa (PSD-95), also known as synapse-associated protein 90 kDa (SAP90), is a membrane-associated protein found in the postsynaptic density of forebrain neurons and certain presynaptic structures in the cerebellum.
PSD-95 contains three PDZ domains, an SH3 domain, and a guanylate kinase-like domain. It plays a critical role in clustering proteins such as NMDA receptors and Shaker-type potassium channels at the synaptic membrane in CNS neurons.
PSD-95 interacts with other proteins through two main mechanisms:
- NMDA receptors and Shaker-type potassium channels share a C-terminal motif (T/SXV) that binds to PDZ domains of PSD-95.
- Neuronal nitric oxide synthase (nNOS), which lacks this motif but has its own PDZ domain, associates with PSD-95 via PDZ-PDZ interactions.
