Target Information
Ubiquitin is a 76 amino acid highly conserved eukaryotic polypeptide that selectively marks cellular proteins for proteolytic degradation by the 26S proteasome. This process regulates the concentrations of key regulatory proteins by limiting their lifespans. Polyubiquitination is a common feature of this modification. Steps include activation of ubiquitin, ATP-dependent formation of a thioester bond with E1 enzyme, transfer to E2 enzyme, and covalent linkage to target proteins directly or via E3 ligase. Deubiquitination enzymes can reverse this process. Ubiquitination is involved in various cellular processes such as cell cycle, growth, differentiation, apoptosis, signal transduction, DNA repair, transcription regulation, and immune responses.
