Target Information

TNPO2 probably functions in nuclear protein import as a nuclear transport receptor. It serves as a receptor for nuclear localization signals (NLS) in cargo substrates. TNPO2 mediates docking of the importin/substrate complex to the nuclear pore complex (NPC) through binding to nucleoporin, followed by energy-dependent translocation through the pore via a Ran-dependent mechanism.
At the nucleoplasmic side of the NPC, Ran binds to the importin, causing dissociation of the importin/substrate complex. Importin is then re-exported from the nucleus to the cytoplasm, where GTP hydrolysis releases Ran. The directionality of nuclear import is conferred by the asymmetric distribution of GTP- and GDP-bound Ran forms between cytoplasm and nucleus.
