Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 regulates transcription of genes such as p21waf1 involved in G1 or G2/M arrest.

The protein structure includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerization domain
  • C-terminal regulatory domain

Phosphorylation of Ser15 is critical for activation and stabilization. Ser392 phosphorylation is associated with tumor formation. p53 plays roles in aging, oxidative stress, and memory processes. It is typically low in normal cells but elevated in transformed cell lines, contributing to malignancy. Mutations in TP53 disrupt DNA binding and tumor suppressor function, observed in many cancers and hereditary conditions like Li-Fraumeni syndrome.
