Target Information
CD11a (LFA-1 alpha, ITGAL) forms the LFA-1 complex with CD18 (alphaLbeta2 integrin). It is crucial for leukocyte adhesion and costimulatory signaling via ICAM-1, -2, and -3.
CD11a expression on leukocytes transitions from low- to high-affinity conformation upon stimulation, enabling strong ligand binding.
It plays a role in immune cell interaction, differentiation, proliferation, and T-cell effector functions.
Blocking CD11a is a therapeutic strategy for inflammatory diseases. Humanized anti-CD11a antibody Efalizumab (Raptiva) and simvastatin binding to CD11a are examples of immunomodulatory mechanisms.
Two transcript variants encoding different isoforms have been identified.
Associated diseases include benign cephalic histiocytosis and leukocyte adhesion deficiency.
