Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation. p53 activates transcription of genes such as p21waf1, leading to G1 or G2/M arrest.
The structure of p53 includes:
- N-terminal transactivation domain
- Central DNA-binding domain
- Oligomerisation domain
- C-terminal regulatory domain
Phosphorylation at Ser15 is critical for activation and stabilization. Phosphorylation at Ser392 is associated with tumor formation. p53 also plays roles in aging, oxidative stress, and cognitive function. It is expressed at low levels in normal cells but highly in transformed cell lines, contributing to malignancy. Mutations in TP53 gene are common in cancers and can lead to loss of tumor suppressor activity. Germline mutations cause cancer-prone syndromes such as Li-Fraumeni syndrome.
