Target Information
STAT3 (Signal Transducer and Activator of Transcription 3) is a member of the STAT protein family. It is phosphorylated by receptor-associated kinases, translocates to the nucleus, and functions as a transcription factor in response to cytokines and growth factors. Coactivators such as CREB-binding protein are required for transcriptional activation by STAT3.
STAT3 can be activated by Interferon-alpha, Interferon-gamma, EGF, PDGF, and IL6. Phosphorylation on tyrosine 705 by JAK1 and JAK2 is essential for STAT3 dimer formation, nuclear translocation, and DNA binding. The STAT3 gene is located on chromosome 17q in humans. It plays a key role in processes such as cell growth and apoptosis. Rac1 regulates STAT3 activity, while PIAS3 acts as a specific inhibitor. Three alternatively spliced transcript variants encoding distinct isoforms have been identified. Deletion of the STAT3 gene in knockout mice is embryonically lethal.
