Product Specific Information

Description:
The 1B1 monoclonal antibody reacts with mouse CD1d, a 48 kDa glycoprotein structurally homologous to MHC class I molecules. CD1d associates with β2-microglobulin and functionally resembles MHC class II. The 1B1 antibody detects CD1d at varying levels on mouse leukocytes and recognizes β2m-associated CD1d.

Applications Tested:

• Tested by flow cytometric analysis of mouse splenocytes.
• Recommended concentration ≤ 0.8 µg per test (100 µL final volume, 10⁵–10⁸ cells/test).
• Antibody should be titrated for optimal performance.

Usage Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability testing.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Technology:

• NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread.
• Excitation: 552 nm; Emission: 755 nm; Laser: 561 nm (Yellow).
• Internal testing shows non-specific staining of B cells in SJL mice; not observed in BALB/c or C57BL/6 strains.

Target Information

CD1d is a heavy chain associated with β2-microglobulin on cortical thymocytes. It can also be expressed independently on human intestinal epithelial cells. CD1d belongs to the CD1 family of MHC-like glycoproteins, mediating the presentation of lipid and glycolipid antigens to T cells.
The human genome contains five CD1 genes clustered on chromosome 1, differing in localization and ligand specificity. CD1d localizes to late endosomes and lysosomes via a tyrosine-based motif.
Diseases associated with CD1D include Mycobacterium malmoense and autoimmune disorders.
