Target Information
In mammals, several isoforms of p21-Activated Protein Kinases (PAKs) are identified:
- PAK-1 (alpha-PAK) and PAK-3 (beta-PAK) are mostly brain-specific.
- PAK-2 (gamma-PAK) is expressed ubiquitously.
Mutations in the gene coding for PAK-3 are associated with X-linked mental retardation. PAK-3 regulates synapse formation and plasticity in the hippocampus and plays key roles in cell shape, motility, and cell death. Autophosphorylation of Thr402 is essential for PAK activation.
