The G-protein-coupled receptor (GPCR) superfamily comprises approximately 600–1,000 members and represents the largest known class of molecular targets with proven therapeutic value.
GPR26 is a brain-specific orphan GPCR exhibiting constitutive activity mediated by G(s)-alpha protein, which stimulates adenylate cyclase and increases intracellular cAMP levels.
- Highest expression: amygdala, hippocampus, thalamus
- Weak expression: testis
- Down-regulated in glioblastoma
- GPR26 depletion increases fat storage in C. elegans
- Deficiency in hypothalamus linked to obesity susceptibility and metabolic disorders (glucose intolerance, hyperinsulinemia, dyslipidemia)
