Twist1/Twist2는 basic helix-loop-helix (bHLH) 전사 인자를 암호화하며, 배아 발생 과정에서 중요한 역할을 합니다. 이 단백질은 DNA의 E-box 서열에 결합하여 두개봉합 형성, 신경관 폐쇄, 사지 발달, 갈색 지방 대사와 관련된 유전자 발현을 조절합니다.
Twist1/Twist2는 여러 암에서 과메틸화 및 과발현되어 종양 세포의 침윤과 전이를 촉진합니다.
또한, 이 유전자의 돌연변이는 Saethre-Chotzen 증후군(두개골 조기융합, 안검하수, 안구격리증)을 유발합니다.
