Target Information
GAPDH (Glyceraldehyde-3-phosphate dehydrogenase)는 해당과정(glycolysis)에 관여하는 촉매 효소로, 동일한 37-kDa 소단위체 4개로 구성된 사량체 형태로 존재합니다. NADPH 존재하에서 1,3-bisphosphoglycerate를 glyceraldehyde 3-phosphate로 가역 환원시키는 반응을 촉매합니다.
GAPDH는 해당과정 외에도 DNA 복제, DNA 수선, 핵 RNA 수출, 막 융합 및 미세소관 결합 등 다양한 세포 과정에 관여합니다. 또한 세포 노화 및 신경퇴행성 질환 관련 세포 표현형, 세포사멸(apoptosis), 암세포 내 신경세포 사멸 등에도 중요한 역할을 합니다.
GAPDH는 amyloid precursor protein, Huntingtin 단백질 등 다양한 단백질과 결합한다고 알려져 있으며, Alzheimer’s disease 및 Huntington’s disease와의 연관성이 보고되었습니다. 또한 actin 및 tubulin과의 상호작용도 보고되어 있습니다.
GAPDH는 대부분의 조직에서 높은 수준으로 발현되므로 Western blot 분석에서 단백질 로딩 컨트롤로 널리 사용됩니다.
