Target Information
STK3 (MST2)는 MAP kinase 신호전달 경로 상류에서 작용하는 STE kinase 계열 단백질로, 세포자멸사 유도 자극(staurosporine, FAS ligand)에 의해 활성화됩니다. Caspase 절단 후 핵으로 이동하여 염색질 응축 및 DNA 절편화를 유도하는 스트레스 반응성, 세포자멸성 키나아제입니다.
Hippo 신호전달 경로의 핵심 구성 요소로, 세포 증식 억제와 세포자멸 촉진을 통해 장기 크기 조절 및 종양 억제에 중요한 역할을 합니다. STK3/MST2와 STK4/MST1은 SAV1과 복합체를 이루어 LATS1/2를 인산화 및 활성화하고, LATS1/2는 YAP1 및 WWTR1/TAZ를 인산화하여 핵 내 이동을 억제함으로써 세포 증식, 사멸, 이동 관련 유전자 발현을 조절합니다.
또한 STK3/MST2와 STK4/MST1은 성숙 간세포의 증식을 억제하고, 간 성체 줄기세포의 활성화 및 종양 형성을 방지하는 데 필요합니다.
