Target Information

DISC1 (Disrupted in Schizophrenia 1) is a multifunctional protein that interacts with multiple proteins of the centrosome and cytoskeletal system. It plays a crucial role in neuronal functions dependent on proper cytoskeletal regulation, including neuronal migration, neurite architecture, and intracellular transport.

Regions of the brain expressing DISC1 include:

  • Hippocampus
  • Lateral septum
  • Amygdala
  • Cerebral cortex
  • Cerebellum
  • Paraventricular hypothalamus

Loss of DISC1 function results in differences in structural organization. The DISC1 gene locus is associated with schizophrenia due to chromosomal translocations. DISC1 encodes a large protein with a globular N-terminal domain and a helical C-terminal domain, both capable of forming interactions with other proteins.

DISC1 interacts with:

  • Centrosome and cytoskeletal proteins: MIPT3, MAP1A, NUDEL
  • Membrane receptor localization proteins: alpha-actinin 2, beta 4-spectrin
  • Signal transduction proteins: ATF-4, ATF-5

Decreased expression of DISC1 in neurons accelerates neuronal integration, leading to aberrant morphological development. This suggests DISC1’s role in dendritic development and synapse formation. DISC1 is thus implicated in intracellular transport, neurite architecture, and neuronal migration, all of which may be pathogenic in the schizophrenic brain.
