Target Information
PTEN was identified as a tumor suppressor frequently mutated in various cancers. The PTEN gene encodes a phosphatidylinositol-3,4,5-trisphosphate 3-phosphatase containing a tensin-like domain and a catalytic domain similar to dual specificity protein tyrosine phosphatases.
Unlike most tyrosine phosphatases, PTEN preferentially dephosphorylates phosphoinositide substrates, negatively regulating intracellular levels of phosphatidylinositol-3,4,5-trisphosphate and the AKT/PKB signaling pathway.
PTEN modulates the AKT-mTOR signaling pathway, influencing neuron positioning, dendritic development, and synapse formation during adult neurogenesis. It may also negatively regulate insulin signaling and glucose metabolism in adipose tissue.
The nuclear monoubiquitinated form shows greater apoptotic potential, while the cytoplasmic nonubiquitinated form has reduced tumor suppressive ability.
