The V3GYWCH monoclonal antibody reacts with human and mouse interferon regulatory factor 8 (IRF8; ICSBP).
IRF8 is a 50 kDa transcription factor essential for the development and differentiation of dendritic cells (DC), particularly plasmacytoid DC (pDC) and CD8α+ DC.
IRF8 deficiency leads to loss of these DC populations and functional impairment of CD8α− DCs. Retroviral IRF8 transduction can restore pDC development from DC progenitors in IRF8-deficient mice.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on normal human peripheral blood cells.
Recommended use ≤ 1 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
Titration is advised for optimal performance.

eFluor 450 is an alternative to Pacific Blue, excited at 405 nm and emits at 446 nm. Ensure instrument compatibility.

Target Information
IRF8 (Interferon Regulatory Factor 8) acts as a transcriptional activator or repressor, binding to regulatory regions of type I IFN and MHC class I genes.
It plays a negative regulatory role in immune cells and is involved in CD8+ dendritic cell differentiation through complex formation with BATF-JUNB heterodimer, recognizing AICE sequences for gene activation.
Mutations in IRF8 can lead to immunodeficiency 32A and 32B.
