Product Specific Information

The FNLAP monoclonal antibody reacts with human latency associated peptide (LAP, pro-TGF beta 1, LAP/TGF beta 1).
TGF beta mediates effects on proliferation, differentiation, and function of many cell types. It is synthesized as a precursor containing LAP at the N-terminus and mature TGF beta at the C-terminus. Processing between amino acids 278 and 279 forms LAP and TGF beta dimers that associate to form the small latent TGF beta complex.

LAP can be secreted or expressed on platelets and activated regulatory T cells, likely through binding to GARP (LRRC32).

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of stimulated normal human peripheral blood cells. Use at 5 µL (0.5 µg) per test in 100 µL final volume.

Fluorophore Information:
eFluor® 450 is an alternative to Pacific Blue®. Emits at 445 nm, excited at 405 nm (Violet laser). Ensure instrument compatibility.

Filtration: 0.2 µm post-manufacturing filtered.


Target Information

TGF beta-1 is a member of the transforming growth factor beta superfamily of cytokines, found ubiquitously in tissues.
It is stored in the extracellular matrix as a latent complex with its prodomain. Activation requires binding of αV integrin to the RGD sequence in the prodomain and mechanical force exertion.
Latent forms consist of TGF-beta, TGF-LAP (latency associated peptide), and LTBP (latent TGF-beta binding protein).
