Product Specific Information

Description:
This MRRBY monoclonal antibody recognizes human and mouse mTOR (mammalian target of rapamycin) when phosphorylated on Ser2448. mTOR is a serine/threonine kinase that regulates cell growth, proliferation, motility, survival, protein synthesis, and transcription in response to nutrient and energy status.

Activated mTOR increases glycolytic enzyme production and nutrient uptake, supporting cell proliferation. It transmits positive signals to p70 S6 kinase and regulates 4E-BP1 activity. Phosphorylation at Ser2448 occurs via the PI3K/Akt pathway, with autophosphorylation at Ser2481. Due to its key role in tumorigenesis, mTOR is a target for anti-cancer therapy.

Applications Reported:
Reported for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-titrated and tested for intracellular staining of stimulated normal human peripheral blood cells. Use at 5 µL (0.06 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Staining Protocols:

  • Protocol A (Two-step, cytoplasmic): Best for simultaneous detection of surface and intracellular proteins.
  • Protocol B (One-step, nuclear): Recommended for transcription factors with surface and phosphorylated proteins.
  • Protocol C (Fixation/Methanol): Provides best phospho-signal discrimination but limited for surface staining.

FRAP1 is the target of rapamycin and its analogues, used as immunosuppressants and cancer therapeutics. Mutations can cause Smith-Kingsmore syndrome.
