Product Specific Information
The MP6-XT22 antibody reacts with mouse tumor necrosis factor-alpha (TNF alpha), a 17 kDa cytokine produced by monocytes, macrophages, neutrophils, NK cells, and CD4(+) T cells. TNF alpha has cytolytic activity against a range of tumor cells and is important in immune regulation. It forms dimers and trimers and also exists as a 26 kDa membrane-bound form.
Applications Reported:
This antibody has been reported for use in intracellular staining followed by flow cytometric analysis.
Applications Tested:
Validated for intracellular staining and flow cytometric analysis of restimulated mouse splenocytes. Recommended usage ≤ 0.25 µg per test (100 µL final volume). Cell number may vary (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.
Fluorophore Information:
eFluor® 450 is an alternative to Pacific Blue®, emitting at 445 nm and excited by the 405 nm violet laser. Ensure instrument compatibility.

Target Information
TNF alpha is a multifunctional proinflammatory cytokine of the TNF superfamily, secreted mainly by macrophages. It binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR receptors and regulates immune cells, proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. TNF alpha forms multimers (dimers, trimers, or pentamers) and shows a single 17 kDa band in SDS-PAGE under non-reducing conditions. Mouse knockout studies indicate neuroprotective roles and tumor necrosis induction. TNF alpha contributes to immune responses against bacterial, viral, parasitic, and fungal infections and has been implicated in autoimmune diseases, insulin resistance, and cancer.
