Description:
The MAb11 antibody reacts with human tumor necrosis factor-alpha (TNF alpha), a 17 kDa cytokine produced by monocytes, macrophages, neutrophils, NK cells, and CD4+ T cells. TNF alpha has cytolytic activity against a range of tumor cells and plays a key role in immune regulation. It forms dimers and trimers and also exists as a 26 kDa membrane-bound form. The MAb11 antibody is a neutralizing antibody.

Applications Reported:
This antibody has been reported for use in intracellular staining followed by flow cytometric analysis.

Applications Tested:
This antibody has been pre-titrated and tested by intracellular staining and flow cytometric analysis of stimulated normal human peripheral blood cells. It can be used at 5 µL (0.5 µg) per test. A test is defined as the amount of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10⁵ to 10⁸ cells/test.

Fluorochrome Information:
eFluor® 450 is an alternative to Pacific Blue®. It emits at 445 nm and is excited with the Violet laser (405 nm). Ensure your instrument is capable of detecting this fluorochrome.

Target Information

TNF alpha is a multifunctional proinflammatory cytokine of the TNF superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cells, proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. It exists as multimers (dimers, trimers, pentamers) but appears as a single 17 kDa band under non-reducing SDS-PAGE.
Knockout studies in mice suggest TNF alpha’s neuroprotective function and tumor necrosis activity. It plays roles in immune responses to bacterial, viral, parasitic, and fungal infections, and in tumor cell cytolysis. TNF alpha is implicated in autoimmune diseases, insulin resistance, and cancer.
