• For IHC (PFA fixed): Incubate antibody for 30 minutes at room temperature.
• For formalin-fixed tissues: Heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, then cool at room temperature for 20 minutes.

Target Information

Interleukin-1 alpha (IL-1α) is a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. It is encoded by the IL1A gene and signals through IL-1RI and IL-1RII, shared with IL-1β.
IL1A is located on chromosome 2q13 and plays a key role in innate host defense by inducing other proinflammatory cytokines and acute-phase responses.
IL-1α activity is regulated by IL-1 Receptor Antagonist (IL-1RA), which competitively inhibits receptor binding.
It participates in immune responses, inflammation, and hematopoiesis, and is produced as a proprotein that is processed and released upon cell injury, inducing apoptosis.
The IL1A gene belongs to a cytokine gene cluster on chromosome 2 and regulates NF-κB and MAP kinase pathways.
IL-1α enhances IL6 and COX-2 expression and nitric oxide (NO) production. Elevated IL-1α levels are associated with chronic inflammatory diseases such as rheumatoid arthritis, psoriasis, and multiple sclerosis.
