Target Information

The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH).
It is commonly expressed at either the N- or C-terminal of recombinant proteins to facilitate isolation or purification via immobilized metal affinity chromatography (IMAC).

Epitope tagging enables detection of recombinant proteins by fusing a known epitope to the target protein, allowing identification even when no specific antibody exists.
His-tagged proteins bind strongly and selectively to Ni²⁺ ions, enabling purification by Ni²⁺-NTA affinity chromatography.
His tag-specific antibodies are widely used for detection and co-immunoprecipitation of tagged proteins.
