A recommended positive control tissue is the cerebellum, though other tissues may also be suitable.

The primary antibody is intended for laboratory professional use to detect the corresponding protein in formalin-fixed, paraffin-embedded tissue for manual qualitative IHC testing. It is used after the primary diagnosis of tumors made through conventional histopathology.

NeuN antibody specifically recognizes the DNA-binding, neuron-specific protein NeuN, present in most CNS and PNS neuronal cell types across vertebrates. NeuN localization is largely restricted to neuronal nuclei and proximal processes. Some neurons, such as INL retinal cells, Cajal-Retzius cells, Purkinje cells, inferior olivary and dentate nucleus neurons, and sympathetic ganglion cells, may not be recognized by NeuN.

Immunoreactivity appears around E9.5 in mouse neural tube development and becomes widespread by E12.5. Strong nuclear staining suggests a regulatory nuclear protein function, though its cytoplasmic role remains unclear. No difference has been observed between protein isolated from nuclei and whole brain extract on immunoblots.

Antibody is used with formalin-fixed and paraffin-embedded sections. Pretreatment with heat-induced or enzymatic epitope retrieval is recommended. IHC staining enables visualization of antigens using sequential antibody applications, enzyme complexes, and chromogenic substrates. The resulting visible reaction is interpreted under a light microscope.

A positive tissue control must be included in each staining procedure. Control tissues should contain both positive and negative staining regions. External positive control materials should be fresh, fixed, processed, and embedded promptly. Weak positive controls are preferred for detecting subtle reagent instability or methodological issues.

Negative control tissues (internal or external) help verify specificity and background staining. If specific staining occurs in negative controls, patient specimen results are invalid.

Target Information

Fox-3 (also known as NEUN, HRNBP3, RBFOX3, or Fox-1 homolog C) is a 350 amino acid protein containing one RNA recognition motif (RRM) domain. It localizes to both the nucleus and cytoplasm and is involved in regulating alternative splicing events.

Fox-3 is encoded on human chromosome 17, which houses over 1,200 genes (~2.5% of the genome). Two major tumor suppressor genes on chromosome 17, p53 and BRCA1, are critical for DNA repair and tumor suppression. Malfunction or loss of p53 is linked to malignant growth and Li-Fraumeni syndrome, while BRCA1 mutations are associated with early-onset breast and other cancers.
