Target Information
LCK는 Src-family tyrosine kinase에 속하며, T-cell receptor (TCR) 복합체를 통한 신호전달에 필수적인 역할을 합니다.
TCR 자극 시, LCK는 zeta 소단위의 ITAM 모티프를 인산화하여 ZAP70의 SH2 도메인 결합 부위를 형성하고, ZAP70을 활성화하여 LAT 어댑터 단백질의 인산화를 통해 후속 신호 플랫폼을 생성합니다.
대부분의 LCK는 세포막에 위치하지만, 일부는 골지체와 연관되어 약한 TCR 자극 시 Raf 활성화에 기여할 수 있습니다. LCK는 N-말단의 myristylation 및 palmitylation 부위, PTK 도메인, SH2 및 SH3 도메인을 포함하며, 이는 각각 인산화티로신 함유 및 프롤린 풍부 모티프와의 단백질-단백질 상호작용을 매개합니다.
또한 LCK는 다양한 자극에 의해 유도되는 세포사멸 조절에 관여하지만, death receptor에 의한 세포사멸에는 관여하지 않습니다.
LCK 관련 질환으로는 Immunodeficiency 22 및 CD45 deficiency가 있으며, 여러 대체 스플라이싱 변이체가 보고되어 있습니다.
