Target Information

IkB-alpha is a 40 kDa protein that inhibits NF-κB activity through protein–protein interactions in the cytosol. Binding of IkB-alpha to NF-κB masks its nuclear localization sequence, preventing nuclear translocation. Various stimuli—such as TNF-α, IL-1β, chemokines, PMA, growth factors, LPS, UV irradiation, and viral infection—activate NF-κB by promoting IkB-alpha degradation.
In humans, the gene is located on chromosome 14q. Activation of NF-κB requires phosphorylation of IkB on serine residues (Ser32 and Ser36) by IkB kinase alpha (IKKα), leading to its degradation. IKKα, along with IKKβ and IKKγ, forms the functional IKK complex, essential for NF-κB activation in response to proinflammatory cytokines.
