Target Information

Histone H2A.X (H2AX) is part of the histone H2A family, one of the four core histones forming the nucleosome core particle.
DNA double-strand breaks (DSBs) induce phosphorylation of serine 139 at the C-terminus of H2AX, producing γ-H2AX, a key DNA damage marker detectable by Western blotting or immunofluorescence.

Phosphorylation is mediated by phosphatidylinositol 3-kinases, potentially including ATM, DNA-PK, and ATR.
H2A.X consists of 143 amino acids and is synthesized in both G1 and S phases. Its mRNA exists in two forms:

  • ~1600 bases with polyA tail (basal type)
  • ~575 bases without polyA tail (replication type)

Histone H2A.X maps to the 11q23.2–q23.3 region of the human chromosome and contributes to chromatin structure and DNA organization.
It also regulates MDC1, a mediator of DNA damage checkpoint protein, at DNA damage sites.
