Product Specific Information
This antibody recognizes the ~90 kDa MDM2 protein and also isoforms at ~57 and ~74/76 kDa.
The epitope recognized by this antibody is within amino acids 26–169 of the human protein.
Positive controls: OsA-CL, MCF-7, HeLa cell lysates
Negative control: HOS cells
Staining of formalin-fixed, paraffin-embedded tissue requires heat-induced epitope retrieval pretreatment.
Target Information
MDM2 is a ubiquitin ligase for p53 and plays a central role in regulating the stability of p53.
Located on chromosome 12q, MDM2 phosphorylation at Ser166 and Ser186 enhances interaction with p300, facilitating ubiquitination and degradation of p53.
MDM2 also inhibits p53 transactivation and can reduce its tumor suppressor function through degradation.
It is overexpressed in various human malignancies including soft tissue carcinomas and breast cancer.
Additionally, MDM2 interacts with proteins such as retinoblastoma 1 and ribosomal protein L5, participating in cell cycle, apoptosis, and tumorigenesis.
More than 40 alternatively spliced transcript variants of MDM2 have been identified in tumor and normal tissues.
