Target Information
PTEN은 다양한 암에서 고빈도로 변이되는 종양 억제 유전자로, phosphatidylinositol-3,4,5-trisphosphate 3-phosphatase 활성을 갖습니다. 이 단백질은 tensin 유사 도메인과 이중 특이성 단백질 티로신 포스파테이스와 유사한 촉매 도메인을 포함합니다.
PTEN은 phosphoinositide 기질을 탈인산화하여 세포 내 phosphatidylinositol-3,4,5-trisphosphate 농도를 음성 조절하고, AKT/PKB 신호 경로를 억제함으로써 종양 억제 기능을 수행합니다.
또한 PTEN은 성체 신경발생 과정에서 뉴런의 위치, 수상돌기 발달 및 시냅스 형성을 조절하는 AKT-mTOR 신호전달 경로의 핵심 조절자로 작용하며, 지방 조직에서 인슐린 신호 및 포도당 대사 조절에도 관여합니다.
핵 내 단일 유비퀴틴화된 형태는 세포사멸 유도 능력이 높으며, 세포질 내 비유비퀴틴화된 형태는 상대적으로 낮은 종양 억제 활성을 보입니다.
