Product Specific Information

The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein belonging to the TNFR family. CD40 is expressed by B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells. It regulates B cell development and maturation, induces Ig isotype-switching, and protects B cells from apoptosis while promoting proliferation.
Interaction of CD40 with CD154 (gp39) on T cells is crucial for T–B cell crosstalk and immune regulation.
5C3 is used for activation of B cells in in vitro functional assays.

Applications Tested: Pre-titrated and validated on normal human peripheral blood cells at 5 µL (0.125 µg) per test (100 µL final volume).
Cell number may range from 10⁵ to 10⁸ cells/test.

eFluor® 450 is an alternative to Pacific Blue®, excited at 405 nm and emits at 445 nm. Ensure instrument compatibility with Violet Laser detection.

Target Information

CD40 antigen is a single-chain glycoprotein, member of the TNF/NGF superfamily, homologous to CD30. Present on all B cells except plasma cells, and on epithelial cells, carcinomas, and lymphoid dendritic cells.
It mediates immune and inflammatory responses, including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.
AT-hook transcription factor AKNA regulates CD40 expression, and TNFR2 acts as a signal transduction adaptor.
CD40–CD154 interaction is implicated in amyloid-beta-induced microglial activation, relevant to Alzheimer’s disease pathogenesis.
Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
Diseases associated with CD40 dysfunction include Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
