Product Specific Information

The 4H11 monoclonal antibody reacts with human CD34, also known as mucosialin. CD34 belongs to a protein family that includes endoglycan and podocalyxin. These are single-pass transmembrane proteins with heavily glycosylated extracellular mucin domains.
CD34 was first identified on hematopoietic progenitors and is widely used as a marker to isolate cells capable of hematopoietic engraftment. It is also expressed on mesenchymal stem cells, tumor cell lines, and vascular endothelia in adults.

Epitopes of CD34 are divided into three classes (I, II, III) based on enzymatic sensitivity. The 4H11 antibody belongs to class III, reacting with a protein epitope.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry of normal human peripheral blood cells. Recommended usage: 5 µL (1 µg) per test in 100 µL final volume.

Target Information

CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) expressed on many stem cell populations. It serves as a stem cell marker, involved in receptor-mediated endocytosis and regulation of adhesion, differentiation, and proliferation of hematopoietic progenitors.
CD34 expression may represent a specific developmental state of hematopoietic cells with altered adhesion and differentiation properties. It may mediate attachment to bone marrow matrix or stromal cells and act as a scaffold for glycan binding to selectins.
The intracellular domain is phosphorylated by protein kinase C, suggesting a role in signal transduction.
Diseases associated with CD34 dysfunction include dermatofibrosarcoma and neurofibroma.
