Target Information
Human immunodeficiency virus (HIV) and related viruses require coreceptors, in addition to CD4, to infect target cells. Among G protein-coupled receptors, CCR5 (CC-CKR-5) is a principal coreceptor for macrophage- and dual-tropic HIV-1 strains, facilitating fusion and entry into human white blood cells. CCR5 is essential for infection by HIV-1, HIV-2, and SIV. Beta-chemokines such as RANTES, MIP-α, and MIP-β act as ligands for CCR5 and can inhibit infection by M-tropic HIV-1. The CCR5 receptor associates with the CD4-gp120 complex of HIV, leading to membrane fusion and viral entry. Its amino-terminal domain and extracellular loops serve as HIV binding sites. CCR5 mRNA is expressed in lymphoid organs and monocytes.
