Description:
The C398.4A monoclonal antibody reacts with ICOS (Inducible Costimulatory molecule, also known as H4/CRP-1/AILIM) and cross-reacts with mouse and rat ICOS. ICOS is a T cell–specific activation molecule, a third member of the CD28/CTLA-4 family, expressed on activated T cells as a 47–57 kDa dimer. It provides potent costimulatory activity for T cell activation and proliferation and is essential for humoral immune response. ICOS binds to its ligand B7h/B7RP-1 on activated APCs, including B cells, and plays a role in autoimmune disease modulation and Th2 cell development.

C398.4A displays strong costimulation of T cell clone proliferation, intermediate costimulation on activated T cells, and weak costimulation on resting T cells. It is a good costimulator of IL-10 production, especially from pre-activated cells expressing high levels of ICOS. The epitope recognized by C398.4A differs from that bound by ICOSL and does not block ICOSL binding.

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of Con A-stimulated mouse splenocytes. Recommended at ≤ 0.06 µg/test (100 µL final volume). Optimal titration required.

Light Sensitivity:
PE-Cyanine5 is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples may be stored up to 3 days at 4°C in the dark using:

488–561 nm excitation; 667 nm emission; compatible with Blue, Green, or Yellow-Green lasers.

Filtration:
0.2 µm post-manufacturing filtered.


Target Information

ICOS (CD278):
Encodes a cell-surface receptor of the CD28/CTLA-4 family. Functions as a homodimer involved in cell-cell signaling, immune response regulation, and T cell proliferation. Mutations are associated with Common Variable Immunodeficiency disorders.
