Product Specific Information

This 22F6 monoclonal antibody reacts with human, mouse, and canine Helios.
Helios is a zinc-finger transcription factor similar to Ikaros, expressed in hematopoietic stem cells of embryos and adult mice. It shows highest expression in regulatory T cells but is not expressed in mature B cells, dendritic cells, or myeloid cells.

Helios can form homodimers and heterodimers with other Ikaros family members to regulate transcription. Eight isoforms exist, some with dominant-negative functions, upregulated in lymphoid malignancies such as T-acute lymphoblastic leukemia.

22F6 cross-reacts with rat Helios.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested using mouse lymph node cells with Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00) following “Staining Intracellular Antigens for Flow Cytometry, Protocol B.”
Use at 5 µL (0.06 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.

Light Sensitivity: Tandem dye sensitive to photo-induced oxidation; protect from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in dark.

Excitation/Emission: 488–561 nm / 667 nm
Laser Compatibility: Blue, Green, Yellow-Green lasers

Target Information

This gene encodes a member of the Ikaros family of zinc-finger proteins (Ikaros, Aiolos, Helios).
These hematopoietic-specific transcription factors regulate lymphocyte development.
Helios forms homo- or heterodimers with other family members and functions mainly in early hematopoietic development.
Multiple transcript variants exist, though biological validity of some remains undetermined.
