The P2H3 monoclonal antibody reacts with human CD62E, a 97–115 kDa member of the selectin family. CD62E (E-selectin or ELAM-1) is an adhesion molecule expressed by endothelial cells upon stimulation with cytokines such as TNFα and IL-1β. Its expression mediates leukocyte rolling and the initial interaction of neutrophils with endothelium during inflammation.
This antibody inhibits cellular adhesion to cytokine-activated endothelial cells.

Flow cytometry, immunoprecipitation, and immunohistochemistry.
Applications Tested:
Validated by flow cytometric analysis of TNFα-activated Human Umbilical Vein Endothelial Cells (HUVEC). Recommended ≤1 µg per test (100 µL final volume). Optimal titration should be determined empirically.

Target Information
Leukocytes adhere to the endothelium during extravasation in postcapillary venules and at inflammatory sites. This process involves adhesion receptors from the selectin and immunoglobulin superfamilies. E-selectin (CD62E) is an endothelial-specific selectin induced by proinflammatory cytokines such as IL-1, TNFα, and bacterial components (e.g., LPS) via NFκB signaling. It plays a key role in leukocyte adhesion during inflammation and is associated with vascular endothelial activation.
