Product Specific Information

Description:
The DREG-56 monoclonal antibody reacts with human CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells. It binds glycosylated, fucosylated, and sulfated sialylated glycoproteins including CD34, GlyCAM-1, and MAdCAM-1. These interactions mediate lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) of peripheral lymphoid tissues.

Applications Reported:

• Flow cytometric analysis
• Non-reducing Western blotting
• Immunohistology of frozen tissue sections
• Inhibition of binding to HEV (use Functional Grade DREG-56, Product #16-0629-85)

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤1 µg/test in 100 µL volume. Cell number should be optimized (10⁵–10⁸ cells/test). Antibody should be titrated for best performance.

Target Information

CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor. It includes:

• C-type lectin binding domain
• EGF-like domain
• Two short consensus repeat (SCR) sequences
• Spacer, transmembrane, and cytoplasmic regions

It is constitutively expressed on most leukocytes including lymphocytes (except many memory T cells), monocytes, and polymorphonuclear cells.
