• For immunohistochemistry (PFA-fixed): incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

Granzyme B is a serine protease found in the granules of cytotoxic T cells and NK cells. It plays a key role in inducing apoptosis in target cells through caspase activation and mitochondrial pathways. Granzyme B interacts with the mannose 6-phosphate receptor, functioning as a death receptor during cytotoxic T cell-induced apoptosis. It cleaves caspase-3 to initiate DNA fragmentation and can also activate apoptosis via Bid protein cleavage. Granzymes are stored in specialized lytic granules of cytotoxic lymphocytes and NK cells, with several types identified in mice and fewer in humans.
